Y Chromosome Test (Y-STR)

Y chromosome testing is used to determine if two or more males share the same paternal lineage and provide a very accurate answer. This test can be used for various reasons including to establish a relationship between male siblings, cousins, uncle and nephew as well as for immigration purposes.

Y chromosome testing method

By analysing the Y chromosome pattern this test can identify if the participants are from the same paternal line. The Y chromosome is inherited and passed on from father to son through the generations. Since it has a relatively infrequent mutation rate, it will remain the same through many generations. It is only broken by the birth of a daughter but if a direct male line can be concluded then all males in that direct male lineage will have an identical Y pattern.

Females do not have a Y chromosome and therefore cannot do a Y chromosome test. However, females wishing to establish paternal lineage can do so by asking a male blood relative to take a Y chromosome test for them.

X Chromosome Test

The X chromosome test is done when two females believe they might share the same father. The test will thus be able to assist you to determine whether you are:

  • Full sisters
  • Half sisters
  • Not related at all

In cases where two females have different mothers and want to confirm whether or not they share the same paternal father (thus, they wish to verify whether they are half sister or no sisters at all) they can carry out a motherless X chromosomes test. This means that they only need to submit their DNA samples.

In cases where sisters have the same mother and question whether they share the same biological father (therefore wanting to determine whether they are full or half siblings), we suggest to carry out an X chromosome test with the mother’s DNA sample. The mother’s DNA samples is necessary to map her X chromosome pattern and exclude it from the test, thereby, examining only the alleged father’s X chromosome pattern.

Cost and information about X chromosome testing

If the mother is not willing to be tested or is simply not available, then a full siblings STR test will need to be done.

An X chromosome test is done between siblings only in cases where a paternity DNA test cannot be carried out. In cases in which the alleged father’s DNA sample is available, a paternity test is recommended.
